On s\u2019accorde \u00e0 reconna\u00eetre que les exosomes jouent un r\u00f4le d\u00e9terminant dans de nombreuses maladies, notamment le cancer. En effet, les exosomes sont de tr\u00e8s petits \u00ab\u2009colis\u2009\u00bb qui voyagent entre les cellules et qui contiennent des renseignements sur la cellule d\u2019origine \u2013 une empreinte digitale de la cellule et de la maladie. Ils seraient en mesure de reprogrammer la cellule r\u00e9ceptrice et de la transformer en cellule canc\u00e9reuse, permettant ainsi la diss\u00e9mination du cancer. Les exosomes sont pr\u00e9sents en grand nombre dans le sang. Il devrait donc \u00eatre possible d\u2019intercepter ces \u00ab\u2009colis\u2009\u00bb, d\u2019en d\u00e9coder le contenu et de d\u00e9terminer la cellule d\u2019origine et la cellule r\u00e9ceptrice cibl\u00e9e. Ces renseignements pourraient \u00eatre utilis\u00e9s pour acc\u00e9l\u00e9rer le diagnostic de la maladie et pr\u00e9voir sa progression, dans le but d\u2019orienter le traitement et d\u2019emp\u00eacher la maladie de progresser. Toutefois, les technologies de pointe employ\u00e9es pour l\u2019analyse des exosomes ne sont pas suffisamment sensibles. De ce fait, des milliers, voire des millions d\u2019exosomes doivent \u00eatre regroup\u00e9s et broy\u00e9s avant d\u2019\u00eatre analys\u00e9s. Ainsi, les renseignements sur les exosomes individuels sont perdus et il est impossible faire la distinction entre le signal des cellules malades et celui des cellules saines.<\/p>\n<p>David Juncker, de l\u2019Universit\u00e9 McGill, a mis au point une technologie r\u00e9volutionnaire appel\u00e9e Digital Omics of Single Exosomes (DOSE) permettant d\u2019analyser des millions d\u2019exosomes en m\u00eame temps. Gr\u00e2ce au financement de la premi\u00e8re phase du concours Innovation de rupture en g\u00e9nomique de G\u00e9nome Canada, la faisabilit\u00e9 de la technologie DOSE a \u00e9t\u00e9 valid\u00e9e, ce qui permettra maintenant de mettre au point une technologie de plateforme pour l\u2019analyse \u00e0 haut d\u00e9bit des exosomes uniques. Avec ses partenaires, le chercheur utilisera la technologie DOSE pour \u00e9tudier les m\u00e9tastases h\u00e9patiques des cancers colorectaux, une forme de cancer r\u00e9pandue touchant plus de 13\u2009000 Canadiens par an et dont le taux de mortalit\u00e9 est sup\u00e9rieur \u00e0 70 %. L\u2019\u00e9quipe vise \u00e0 identifier l&rsquo;\u00ab\u2009empreinte digitale\u2009\u00bb exosomique de ces m\u00e9tastases, laquelle pourrait ensuite \u00eatre utilis\u00e9e afin de mieux comprendre ce cancer, de d\u00e9velopper des tests diagnostiques sanguins et d\u2019orienter les interventions th\u00e9rapeutiques.<\/p>\n<p>On pr\u00e9voit que l\u2019\u00e9laboration de la technologie DOSE ouvrira la voie \u00e0 la commercialisation par l\u2019interm\u00e9diaire d\u2019une entreprise d\u00e9riv\u00e9e qui fournira des instruments, des r\u00e9actifs et des services. La technologie DOSE pourrait transformer la recherche sur le cancer, les essais cliniques et la prise en charge globale du cancer, tout cela gr\u00e2ce \u00e0 un simple pr\u00e9l\u00e8vement sanguin non invasif et peu co\u00fbteux.
